Mass Propagation of Betula pendula ¢¥ Trost Dwarf ¢¥ through Axillary Buds in Vitro

Abstract
To establish a rapid micropropagation method, axillary buds of Betula pendula ¢¥Trost Dwarf¢¥ were cultured on the woody plant medium (WPM), broad-leaved trees medium (BTM), Murashige & Skoog¢¥s medium (MS), Gress-hoff & Doy¢¥s medium (GD) supplemented with benzylaminopurine (BAP), Kinetin, naphthalene acetic acid (NAA) and indole butyric acid (IBA). The results are summarized as follows:
Etiolation was required for shoot elongation from axillary buds and resulted in shoot elongation and induction of multiple shoot at the basal part of explants. WPM and BTM containing 0.5 §·/§¤ of BAP were adequate for multiple shoots formation by using etiolated explant. In the combination of media and plant growth regulators, multiple shoot formation and shoot elongation of Betula pendula ¢¥Trost Dwarf was stimulated by WPM supplemented with 0.5 §·/§¤ BAP and 0.01 §·/§¤ NAA or 0.01 §·/§¤ IBA as the average shoot number 9.8 and 10.28, while the number of shoots in WPM without BAP was 1.80. A 100% rooting was achieved on GD and 1/2 GD medium supplemented with 0.2 §·/§¤ IBA. The survival rates after transplanting into a mixture of peat moss and perlite (1:1 v/v) was 95% and then established plantlets were moved into the greenhouse and the survival rate was 95%, too. Conclusively, after 5 weeks of culture, we have obtained over 1000 plants of Betula pendula ¢¥Trost Dwarf 3¢¥ by the culture of 10 axillary buds.
